Statistical genomics for Life-Sience Statistical genomics involves the integration of both statistics and genetics. Identification and interpretation of the genetic basis of human traits is the goal of population association and linkage studies. Association analysis answers the question: what are the patterns of polymorphisms that vary systematically between individuals with different disease states and could therefore represent the effects of risk-enhancing or protective alleles. Linkage analysis is focused on mapping the functionality of genes to the chromosomes, supposing that gene which is responsible for the disease is located close to its marker and they are segregating together within a pedigree. In order to increase the power of existing marker sets in genome-wide association studies, imputation algorithms are used. Linkage analysis can be performed with any number of marker loci, multiple trait loci, and multiple genomics segments with the Markov chain Monte Carlo approach. We aim to identify single-nucleotide polymorphisms (SNPs) that are associated with our previously established brain correlate of greater reserve capacity (i.e. higher left frontal cortex connectivity, Franzmeier et al., 2017, 2018; Neitzel et al., 2019). To this end, we plan to investigate imaging and genetic data of about 22,000 participants of the UK biobank. Imaging methods are preprocessing of fMRI data (mainly image registration and filtering) and connectivity analysis (correlation of timeseries data). The genetic analysis includes a genome-wide association analysis and computation of polygenetic risk scores.
