We will dissect the heterogeneity of cutaneous human T cell subsets with unprecedented breadth and depth using multi-disciplinary state-of-the-art and novel experimental methodologies, computational approaches and clinical settings for human in vivo perturbation. This will provide a molecular fingerprint for distinct T cell populations that will allow us to further investigate their phenotypic and functional identity, their ontogeny and dynamic relationship within the skin and with the recirculating immune compartment. To this end we will bring together pseudotime reconstruction methods and population-dynamic modelling based on single cell transcriptome data. These insights will be essential to understand the orchestration of the tissue resident T cell compartment with crucial implications for first-line protective barrier immunity, chronic inflammatory skin diseases and vaccination strategies.
