Abstract:

Light field microscopy is a newly developed technique that allows to image the whole brain of behaving animals at unprecedented speed (Nature Methods volume 11, pages 727–730, Prevedel et al. 2014). We recently adapted the technique to image the whole brain of behaving flies at 200Hz (https://journals.plos.org/plosbiology/article?id=10.1371/journal.pbio.2006732). However, this technique is quite computationally demanding, as it requires to solve the inverse problem of which volume intensity led to the observed light field image. This requires the use of a GPU cluster to deconvolve the images. Indeed a full experiment of ~10000 images necessitates 15 clusters for about 10h to reconstruct, which would be nearly impossible to do locally. Acquiring data for several flies in different conditions would allow us to understand the network dynamics underlying behavior and internal states in flies.
